The Chitosan-Loaded Nanoparticles with Commercial Vaccine Plusvac® SL Targeting Nano-Immunotherapy for Dust Mite Allergies

Abstract

A nanoimunoterapia apresenta uma alternativa promissora às terapias convencionais para
alergias a ácaros, protegendo e direcionando o ingrediente biologicamente ativo, aumentando assim sua
eficiência. Objetivo: Este estudo investiga a síntese e o corpo das nanopartículas de quitosana
com uma vacina comercial (Plusvac) voltada para nanoimunoterapia. Metodologia: As
nanopartículas foram sintetizadas por meio de gelificação iônica e transmitidas com o conteúdo proteico da
vacina comercial. Resultados: Para uma proporção de síntese de 3:1 (quitosana: poliânion), nanopartículas com
diâmetro médio de 200 nm e potencial zeta de +32,73 mV foram produzidas com sucesso.
A eficiência de carregamento foi de 19,40%. Para uma massa inicial de 627,443 μg de proteína encapsulada, uma
liberação cumulativa de 33,11% (207,0 μg) foi liberada após 60 horas de monitoramento. Conclusão:
As características das nanocápsulas, com alérgenos encapsulados em seu interior, indicam que
este estudo é promissor e relevante para futuras aplicações em administração sublingual para
dessensibilização de indivíduos com alergias atópicas.

Marcel Guimarães Martins, Federal University of Rio de Janeiro

Chemical Engineer, graduated from the Fluminense Federal University (UFF). He carried out research at the Institute of Chemistry - UFF, with an emphasis on Applied Inorganic Synthesis and Molecular Magnetism. He interned at the company PETROBRAS, working on process optimization at the Duque de Caxias refinery (REDUC). He completed an exchange at the University of Granada (UGR), in Spain. Master and PhD in Chemical Engineering from the COPPE Chemical Engineering Program, at the Federal University of Rio de Janeiro (PEQ/COPPE/UFRJ), working in the area of ​​encapsulation of magnetic nanoparticles in a polymeric matrix. During the master's degree, he also acted as a student representative in the program (PEQ/COPPE/UFRJ). Active in the line of nanomagnetism and biotechnology research, developing multifunctional nanosystems for health applications. Founding partner of the startup Magtech Soluções em Nanoparticles Magnetéticas Ltda. ME (@magtechbrasil), which develops solutions in the biotechnology market with a focus on health. He served as PDI-Market Relationship Director for the National Association of Biotechnology and Life Sciences Companies (ANBIOTEC) in the 2021-2022 biennium. He also works as a Specialist in Innovation Projects at Fiocruz (INOVA Fiocruz Program).

Raquel Corrêa Diniz, Universidade Federal do Rio de Janeiro

Studying for a degree in Biological Sciences at the Federal University of Rio de Janeiro - UFRJ. He studied Biotechnology Technician at IOC - FIO CRUZ from March 2020 to September 2022. Carried out Technological Initiation at the company Extractos Allergens MQ at UFRJ from January 2024 to date. Undertook Scientific Initiation at the Microbiology Laboratory of the UFRJ Biology Institute from August 2021 to October 2022 research line: Bacterial genetics and bacteria-host interaction. He has experience in the area of ​​Chemical Engineering, with an emphasis on Automatic Control and development of bioprocesses to obtain organic acids and biofuels. He was a CNPq postdoctoral fellow (2004-2006) in the construction of immunosensors in collaboration with the Postgraduate Program in Biochemistry (Laboratory for the Study of Structure and Function of Proteins UFJF/MG). Postdoctoral (2006 - 2008) in the Postgraduate Program in Chemical Engineering and Process Biochemistry at EQ/UFRJ with biosensors for the detection of phenolic compounds. She is currently Associate Professor IV at the Department of Chemical and Petroleum Engineering at Universidade Federal Fluminense (UFF) since 2010. Coordinator of scientific research and extension at the Bioprocesses and Biotechnology Laboratory (BioPro). Collaborating researcher at the ADDLabs Research Group - Active Documentation and Intelligent Design Laboratory at the UFF Computing Institute (2017 - 2019). Collaborating Researcher of the Professor Nei Pereira Jr Research Group - Bioprocess Development, EQ/UFRJ.
